- 方法一:其制备方法是叔丁醇与硫氰酸铵反应，生成硫氰酸叔丁酯，再在催化剂存在下转位，生成异硫氰酸叔丁酯。反应方程式：(CH 3 ) 3 COH+NH 4 SCN+HCl→[△](CH 3 ) 3 CSCN+NH 4 Cl+H 2 O (CH 3 ) 3 CSCN[催化剂]→[异构化](CH 3 ) 3 CNCS 在反应釜中加入固体硫氰酸铵，随后真空抽入叔丁醇和水。将盐酸泵入盐酸计量滴加槽，搅拌，加热，在75℃下滴加盐酸2～3h，滴完后再保温反应4～5h。然后冷至室温，静置分层。油层水洗2次后抽入中间储槽，分净水后再抽入转位反应釜中，加催化剂，搅拌，在40～50℃反应4h，所得物料进行减压蒸馏，即得到异硫氰酸叔丁酯。
